Carotenogenesis of Staphylococcus aureus: New insights and impact on membrane biophysical properties

Staphyloxanthin (STX) is a saccharolipid derived from carotenoid in Staphylococcus aureus involved oxidative-stress tolerance and antimicrobial peptide resistance. STX influences the biophysical properties of bacterial membrane has been associated to formation lipid domains regulation methicillin-resistance. In this work, targeted metabolomics characterization study was carried out investigate biosynthetic pathways carotenoids, their impact on properties. Five different S. strains were investigated, including three wild-type containing crtM gene related biosynthesis, crtM-deletion mutant, crtMN plasmid-complemented variant. LC-DAD-MS/MS analysis extracts allowed identification 34 metabolites carotenogenesis at growth phases (8, 24 48 h), showing progression these as bacteria advances into stationary phase. For first time, 22 members large family carotenoids identified, STX-homologues, well Dehydro-STX Dehydro-STX-homologues. Moreover, thermotropic behavior CH2 stretch acyl chains live cells by FTIR, show that presence increases chain order temperature.
